Lymphocvte Transformation Induced by A - kutologous Cells : Stimulation by Cultured Lymphoblast Lines MARc E . WEKSLER and GARY BIRNBAUM

چکیده
cultured lymphoblasts required physical contact with responding lymphocytes to induce transformiiation. In quantitative terms, lymphocytes incorporate as much thymidine when, mixed with irradiated cultured lymphoblasts as they (lo in response to l)hytolhenma-glutin1in. The stimulation of lymphocyte transformation by allogeneic cultured lymphoblasts did not parallel the stimulation of lymphocyte transformation by leukocytes from the donor of the lymphoblast culture. The stimulatory determinants on the cultured lymphoblast are unaffected by neuramini(lase but destroyed by trypsin. The trvpsin-treated cultured lymphoblasts regain their capacity to stimulate atutologous lymphocyte transformation witlhin 48 hr in culture. Cultured lymphoblasts l)ossess concanavalin A binding sites. Concanavalin A inhibits the capacity of cultured lymphoblasts to stimulate autologous lymphocyte transformation. The relevance of these findings to EB virus infection of culture(d lymplhoblasts and to imilmune surveillance is discusse(l.

Removal of Mitogen from Leukocytes Exposed to Phytohemagglutinin or
The stimulation of lymphocyte transformation by autologous human lymphoblasts established in continuous culture has been reported by several laboratories (1-6). The nature of the determinants on cultured lymphoblasts that stimulate transformation of autologous lymphocytes is not clear, but may be related to what appears to be a universal infection (latent or expressed) of all lymphoblast lines ...
متن کاملLymphocyte Transformation Induced by Autologous Cells
Lymphocytes incubated with phytohemagglutinin or concanavalin A develop the capacity to stimulate autologous lymphocyte transformation. This is not attributable to residual mitogen contaminating the lymphoblastic cell preparation as: (a) the dissociation of mitogen from the lymphoblastic cell preparation increases the degree of stimulation observed and (b) the kinetics of lymphocyte transformat...
متن کاملEffect of Tribulus Terrestris L. on Expression of ICAM-1, VCAM-1, E-Selectin and Proteome Profile of Human Endothelial Cells In-Vitro
Background: Atherosclerosis is a chronic inflammation that interferes with blood arteries functions due to the accumulation of low density lipids and cholesterol. Objective: To investigate the effect of aqueous extract and saponin fraction of Tribulus terrestris L. (TT) on the proteome and expression of intracellular adhesion molecule-1 (ICAM-1), vascu...
متن کاملA Comparison between the Cytotoxicity Induced by Gossypol in Two Testicular Cell Lines
Background:Gossypol is a yellow toxic pigment from the cottonseed that can cause acute or chronic toxicity in humans and animals by affecting the testicular tissues. Nowadays cottonseed is used as food supplement for ruminants specially the sheep. In this study, two different stem cell lines of testicular tissue including GC1-spg (mouse testis) and SFTF-PI43 (sheep testis) cells were used to ev...
متن کاملReversal of UDP-galactose 4-epimerase deficiency of human leukocytes in culture.
Stimulation with phytohemagglutinin of the leukocytes from six of the seven known individuals with UDP-galactose 4-epimerase (= UDP-glucose 4-epimerase; EC 5.1.3.2) deficiency consistently resulted in the appearance of epimerase activity in the cultured cells. A long-term lymphoblast culture derived from one proband also contained an active epimerase enzyme. A comparison of the properties of th...
